ABSTRACT
Oleanolic acid (OA), a pentacyclic triterpenoid, exhibits potential anti‐tumor activity against many tumor cell lines. This study aims to examine the anti‐tumor activity of OA on pancreatic cancer cells and its potential molecular mechanism. The results showed that the proliferation of Panc‐28 cells was inhibited by OA in a concentration‐dependent manner, with an IC50 (The half maximal inhibitory concentration) value of 46.35 µg ml−1, as determined by MTT (3‐(4,5‐Dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide) assay. The cell cycle was arrested in S phase and G2/M phase by OA. The study also showed that OA could induce remarkable apoptosis, evidenced by an increased percentage of early/late apoptotic cells, DNA ladder and nuclear morphology change. Further study revealed that OA could induce Reactive Oxygen Species (ROS) generation, mitochondrial depolarization, release of cytochrome C, lysosomal membrane permeabilization and leakage of cathepin B. The expression of apoptosis‐correlated proteins was also affected in cells treated with OA, including activation of caspases‐3/9 and cleavage of PARP. Further study confirmed that ROS scavenger vitamin C could reverse the apoptosis induced by OA in Panc‐28 cells. Our results provide evidence that OA arrests the cell cycle and induces apoptosis, possibly via ROS‐mediated mitochondrial and a lysosomal pathway in Panc‐28 cells.

The next day the injection spot turned read and was eatching. Immediatelly I stopped taking Olive Leaf Extract. The other day the red area enlarget as big as the palm of the hand. The therd day I went to the doctor. He was alarmed and adviced me to go to the hospital. I declined and he used a black marker, starting from the shoulder to the elbow, while I was leaving he told me: if it is going to grow bigger than marked area then go to E.R.
In fact it took about 10 days to normlize. I was scared to use Olive Leaf Extract because of this reaction.
After health care system let me to go for good, and the loose stool was persistent, I decided to give another try.
I started taking Spanish Olive Leaf Extract 25% Oleuropein. The result was imminent. I started Low and slow with 1 Capsule per day. After 3 days I encreased to 2 capsules per day. In a week my intestin is normalised. I plan to encrease to 3 capsules per day for some time, and after go to a maintenance dose 2 capsules per day.
Based on my opinion the Olive Leaf Extract is very potent. It can create strong alergic reactions with vacines, and the human body tolerates it well. If used for treatment of different medical condition Start Low and slow but to have effect you must reach a lethal dosage for the pathogen and stay in this level for a while before going down to a maintenance dose.

3.3 EEOL induce apoptosis of pancreatic cancer cells
The effect of EOOS and EEOL on PC cell cycle progression was evaluated (Figure 3A). Overall, the effect of basil extracts on cell cycle progression was not conclusive. In AsPC-1 cells, EOOS decreased the percentage of cells in the G2/M-phase, whereas there was an increase in the S phase and no significant effect was noted on the G1 phase. On the other hand, in MiaPaCa cells, both EOOS and EEOL extracts appeared to promote the progression through the cell cycle as evidenced by a decrease in the percentage of EOOS and EEOL treated cells in the G1 phase and an increase in the percentage in the S and G2/M phases. Nevertheless, the percentage of apoptotic and necrotic cells after basil extract treatment was analyzed after Annexin V-propidium iodide staining and the results revealed the percentage of apoptotic cells was significantly higher upon treatment with EEOL, revealing the cytotoxic mechanism of the extract on PC cells.
3.4 EEOL/EOOS downregulates the expression of proteins that promote pancreatic cancer invasion and metastasis
As we had observed a significant inhibition of motility and invasion of PC cells by EEOL and EOOS, we also sought to investigate whether there was a change in the molecular pathways mediating these functions. For this, AsPC-1 cells were treated with either EEOL or EOOS for 48 hours followed by western blotting and Real time RT-PCR to examine the change in the expression of key proteins and genes. Both EEOL and EOOS significantly decreased the activation of the key mediators of PC cell survival, proliferation, metastasis, invasion and chemoresistance: the extracellular signaling regulated kinase (ERK-1/2) and focal adhesion kinase (FAK) (Figure 4A). Activation of NF-kB, specifically the p65 subunit, is known to be a key activator of pathways that promote proliferation, invasion and metastasis [26]. Both EEOL and EOOS downregulated the protein expression of the p65 subunit of NF-κB in PC cells suggesting that inhibition of NF-kB is one of the possible mechanisms by which basil oil or leaf extract inhibits the aggressive behavior of PC cells (Figure 4A).

Abstract
Pancreatic cancer (PaCa) is one of the most lethal cancers, with an estimated 5-year survival of <5% even when patients are given the best treatment available. In addition, these treatments are often toxic and expensive, thus new agents which are safe, affordable and effective are urgently needed. We describe here the results of our study with acetyl-11-keto-β-boswellic acid (AKBA), an agent obtained from an Ayurvedic medicine, gum resin of Boswellia serrata.Whether AKBA has an activity against human PaCa, was examined in in vitro models and in an orthotopic nude mouse model of PaCa. We found that AKBA inhibited the proliferation of four different PaCa cell lines (AsPC-1, PANC-28, and MIA PaCa-2 with K-Ras and p53 mutations, and BxPC-3 with wild-type K-Ras and p53 mutation). These effects correlated with an inhibition of constitutively active NF-κB and suppression of NF-κB regulating gene expression. AKBA also induced apoptosis, and sensitized the cells to apoptotic effects of gemcitabine. In the orthotopic nude mouse model of PaCa, p.o. administration of AKBA alone (100 mg/kg) significantly inhibited the tumor growth; this activity was enhanced by gemcitabine. In addition, AKBA inhibited the metastasis of the PaCa to spleen, liver, and lungs. This correlated with decreases in Ki-67, a biomarker of proliferation, and CD31, a biomarker of microvessel density, in the tumor tissue. AKBA produced significant decreases in the expression of NF-κB regulating genes in the tissues. Immunohistochemical analysis also showed AKBA downregulated the expression of COX-2, MMP-9, CXCR4, and VEGF in the tissues. Overall these results demonstrate that AKBA can suppress the growth and metastasis of human pancreatic tumors in an orthotopic nude mouse model that correlates with modulation of multiple targets.

This stuff rocks! It 100% works! When I first took it, I was monitoring "what it would do to me." For me, it takes about 45min to an hour before it kicks in and...it just makes me feel good. UA inhibits distant organ metastasis from orthotopically implanted pancreatic cancer in nude mice
At autopsy, the mice in each treatment group were examined for the presence of metastases. The results showed that pancreatic cancer metastasis developed more frequently in the spleen and liver of vehicle-treated mice than in mice treated with UA or gemcitabine alone. UA and gemcitabine respectively decreased to 33.3% and 22.2% metastasis of pancreatic cancer cells in spleen while in liver it decreased to 30% and 35%. Maximum inhibition of metastasis, however, was observed in the group treated with the combination of UA and gemcitabine 66.6% in spleen and 65% in liver (Figure (Figure4A4A).
Please note: I looked into gemcitabine which is a chemotherapy drug, and it appears that THIS STUDY, which found gemcitabine to work better than proleolytic enzymes, was highly biased. Next we examined whether UA modulates the expression of proteins involved in cell survival, invasion, and metastasis in orthotopically implanted pancreatic tumors. Western blotting revealed significant reductions in the expression of markers for cell survival (survivin and Bcl-2), proliferation (cyclin D1), inflammation (COX-2), and metastasis (ICAM-1 and MMP-9) in tumors from the groups treated with UA or gemcitabine alone compared with those from the control group. However, gemcitabine in combination with UA was more effective in suppressing the expression of these proteins than either UA alone or gemcitabine alone (Figure (Figure5C5C).
Immunohistochemical analysis of pancreatic tumor tissues also revealed that UA, alone or in combination with gemcitabine, significantly decreased the expression of cyclin D1, COX-2, VEGF, and MMP-9 molecules compared with the control treatment (Figure (Figure6A6A).

The metastatic spread of tumor cells from the original site is the cause of the high mortality rates associated with cancer. There are at least four interrelated biological events required for tumor metastasis: Angiogenesis, cell adhesion, cell invasion and cell proliferation (52). The interesting aspect of the anticancer activity of bromelain is its inhibitory effect on cancer metastasis. Tumor cells initiate platelet activation, as well as the platelet-based production of multiple factors facilitating angiogenesis. Additionally, tumor cells, to varying degrees, possess the capacity to surround themselves with platelets, forming tumor-platelet aggregates that protect tumor cells from immune recognition (59). Oral administration of bromelain, as assessed by in vitro assays resulted in a reduction of platelet aggregation and activation (60). A previous study demonstrated that in vitro bromelain treatment of platelets from healthy volunteers significantly reduced platelet count (61). The ability of bromelain to inhibit platelet activation is associated with its proteolytic activity (17). After doing all this research on Bromelain, it's definitely going to be in my daily regimen! I think that it's TOTALLY WORTH IT and I would HIGHLY RECOMMEND IT!
